While the majority of patients tolerate radiation and chemotherapy well, only a small percentage of patients suffer from acute side effects which can result in significant and life-threatening complications. There is an urgent need to develop new strategies to prevent radiation and chemotherapy-induced toxicity. To address this need, we are investigating the efficacy of Biopharmaceuticals, including monoclonal antibodies (mAbs), in the prevention of radiation and chemotherapy-induced toxicity. Methods To evaluate the efficacy of mAbs for the prevention of radiation and chemotherapy-induced toxicity, animals were administered a cocktail of mAbs 24h prior to radiation or chemotherapy. Non-specific mAbs, including anti-HER2/neu (22c6), were used in these studies. The effects of radiation on normal tissues, including the intestinal tract, and the effect of radiation and chemotherapy on immune function were evaluated in these studies. Results Anti-HER2/neu mAb treatment significantly decreased the histopathological changes observed in the intestinal tract of mice treated with radiation and chemotherapy. In addition, radiation and chemotherapy caused a significant decrease in natural killer (NK) cell activity. However, anti-HER2/neu mAb treatment restored radiation and chemotherapy-induced NK cell dysfunction. Anti-HER2/neu mAb treatment also significantly decreased the number of radiation and chemotherapy-induced acute graft-vs.-host disease (GVHD) lesions in mice. We are now in the process of evaluating whether treatment with anti-HER2/neu mAb also protects against radiation and chemotherapy-induced late toxicity.
